Vancomycin
Mechanism of action
Inhibits cell wall synthesis BACERICIDAL
( Except Vs streptococci Bacteriostatic )
Antibacterial activity
- Effective mainly vs G+ve bacteria (especially staph)
- Effective on anerobic G+ve
- NOT effective vs G-ve bacteria
- Given with
( streptomycin + gentamycin ) Vs E.faecalis
Pharmacokinetics
- Not absorbed from GIT
given I.V
- Widely distributed
except to CSF
- Only 10 % bound
- Excreted unchanged
in urine
- T1/2 = 8 hours
Clinical uses
- Staph infections not responding to other
anti-biotics
- Serious infections by MRSA and MRSE
- Antibiotics-associated colitis ( ACC ) orally
- Prophylaxis for endocarditis
- prophylaxis for major
surgical procedures
Adverse effects
- GIT upset
- Fever , rashes
- Thrombophilibitis
- Ototoxicity & nephrotoxicity
- Red man
( red neck syndrome)
- Hypersensitivity
